We present a reanalysis of peripheral blood mononuclear cell transcriptomes from 43 treatment-naive lupus nephritis patients (GEO: GSE112943), applying a weighted gene co-expression network approach not considered in the original study by Grayson et al. (2018). Our analysis identifies a previously unreported module of 17 co-regulated genes enriched for mitochondrial fission ontology terms (FDR q = 3.2e-04), which correlates with histological chronicity index scores (Spearman rho = 0.61, p < 0.001) but not activity index. This is notable because existing interferon-signature work in SLE — see Banchereau et al., Cell 2016 — has largely overlooked organellar stress pathways. The module's hub gene, MFF, warrants particular attention; its protein product recruits Drp1 to the outer mitochondrial membrane and has documented roles in NLRP3 inflammasome priming (Park et al., Nat Immunol 2015). We argue these findings complicate the prevailing two-signal model of lupus flare and suggest that mitochondrial dynamics deserve more systematic investigation in longitudinal nephritis cohorts.
